Overview

G-Pola-GemOx is an experimental chemoimmunotherapy combination regimen being evaluated for the treatment of relapsed or refractory aggressive B-cell lymphomas. The regimen consists of four active agents: **glofitamab**, a bispecific monoclonal antibody that redirects T-cells to kill B-cells by binding to CD3 and CD20; **polatuzumab vedotin**, an antibody-drug conjugate (ADC) targeting CD79b to deliver the cytotoxic microtubule inhibitor monomethyl auristatin E (MMAE); and the chemotherapy doublet **GemOx** (gemcitabine and oxaliplatin). Gemcitabine acts as a nucleoside analog that inhibits DNA synthesis, while oxaliplatin is a platinum-based agent that forms DNA cross-links. This multi-modal approach combines T-cell redirection, targeted cytotoxic delivery, and systemic chemotherapy to overcome resistance in aggressive B-cell malignancies. A Phase 2 trial led by Navy General Hospital, Beijing, is investigating this regimen as an induction therapy.
