Overview

This is a multi-agent investigational combination regimen consisting of five drugs with distinct mechanisms of action, primarily under evaluation for the treatment of B-cell lymphomas and related hematologic malignancies. - **Glofitamab** is a bispecific monoclonal antibody targeting CD20 and CD3, designed to redirect T cells to kill malignant B cells. - **Polatuzumab vedotin** is an antibody-drug conjugate (ADC) targeting CD79b on B cells, delivering the cytotoxic agent monomethyl auristatin E (MMAE) directly into malignant cells. - **Zanubrutinib** is a small molecule Bruton’s tyrosine kinase (BTK) inhibitor that blocks B-cell receptor signaling, leading to reduced proliferation and survival of malignant B cells[3][7][9]. - **Lenalidomide** is an immunomodulatory agent that enhances immune cell function and has direct antineoplastic effects[6][8]. - **Prednisone** is a synthetic glucocorticoid with anti-inflammatory and immunosuppressive properties commonly used in lymphoma regimens. This combination aims to leverage synergistic effects from targeted therapy (glofitamab, polatuzumab vedotin, zanubrutinib), immunomodulation (lenalidomide), and corticosteroid activity (prednisone). It has not been established as a standard regimen but reflects ongoing efforts in clinical research for aggressive or refractory forms of lymphoma.
