This combination regimen is a multi-drug chemo-immunotherapy being investigated for the treatment of newly diagnosed diffuse large B-cell lymphoma (DLBCL). It integrates three distinct targeted biological agents with a modified chemotherapy backbone. **Glofitamab** is a bispecific T-cell engager (BiTE) that binds to CD20 on B-cells and CD3 on T-cells, inducing T-cell mediated cytotoxicity. **Polatuzumab vedotin** is an antibody-drug conjugate (ADC) targeting CD79b, delivering the microtubule inhibitor monomethyl auristatin E (MMAE) directly to malignant B-cells. **Zuberitamab** is a recombinant humanized anti-CD20 monoclonal antibody that promotes B-cell depletion through antibody-dependent cellular cytotoxicity (ADCC) and complement-dependent cytotoxicity (CDC). The regimen is often administered in a response-adapted manner, where patients not achieving a complete response (CR) after initial cycles receive the addition of **CHP** (cyclophosphamide, doxorubicin, and prednisone) chemotherapy. This approach aims to maximize efficacy by targeting multiple B-cell antigens and leveraging both immune-mediated and cytotoxic mechanisms of action.
