Drug intelligence / Profile preview
Gloriosine is a colchicine-like natural alkaloid isolated from the roots of *Gloriosa superba* (glory lily). Structurally identified as N-deacetyl-N-formylcolchicine, gloriosine acts as a microtubule-targeting agent that binds to the colchicine-binding site of tubulin, disrupting microtubule polymerization and arresting the cell cycle at the G2/M phase. In addition to its antimitotic properties, gloriosine has been shown to induce autophagic cell death in non-small cell lung cancer (NSCLC) cells by negatively regulating Yes-associated protein (YAP) transcriptional activity, which subsequently suppresses the AKT/mTOR pathway. In triple-negative breast cancer (TNBC) models, gloriosine also downregulates glutaminase 1 (GLS1), suppressing glutamine metabolism and inducing ferroptosis. It is currently being investigated in preclinical research as a potential anticancer therapeutic candidate with lower toxicity to normal cells compared to colchicine.
