Overview

Glovadalen (developmental code name UCB-0022) is an orally active, centrally penetrant small molecule under development for the treatment of Parkinson's disease. It acts as a positive allosteric modulator of the dopamine receptor D1 (D1R), enhancing the receptor’s signaling in the presence of endogenous dopamine without intrinsic agonist activity. By binding outside the dopamine site on D1R, it increases dopamine potency up to 10-fold and shows high selectivity for D1 over other dopamine receptors. The drug aims to improve motor symptoms in Parkinson’s disease while reducing dyskinesia associated with traditional dopaminergic therapies. Glovadalen is being developed by UCB and is currently in Phase 2 clinical trials as an adjunctive therapy to standard-of-care medications such as levodopa[1][2][5][6].
