GLP-ABD-XTEN144 is a **long-acting glucagon-like peptide-1 (GLP-1) receptor agonist** fusion protein engineered for the treatment of type 2 diabetes (T2D). It is designed by fusing a GLP-1 analog to both an albumin-binding domain (ABD) and an XTEN polypeptide sequence (XTEN144) to extend its half-life in vivo. In preclinical studies, GLP-ABD-XTEN144 exhibited stronger albumin binding (Kd 5.50 nM for HSA), a prolonged half-life in mice (12.9 hours), and more pronounced hypoglycemic and appetite-suppressing effects compared to related constructs. This design synergistically exploits both ABD and XTEN approaches to maximize pharmacokinetic improvements, supporting its development for sustained glucose and appetite control in T2D[1][2].
