Overview

GLPG0974 is an orally available small molecule antagonist of the free fatty acid receptor 2 (FFA2, also known as GPR43), a G protein-coupled receptor activated by short-chain fatty acids produced from dietary fiber fermentation in the gut. FFA2 plays a key role in neutrophil activation and migration, contributing to inflammatory processes such as those seen in ulcerative colitis. By selectively inhibiting FFA2, GLPG0974 reduces neutrophil migration and activation, offering a novel anti-inflammatory mechanism for potential treatment of inflammatory bowel diseases. The drug was developed by Galapagos and has undergone Phase 1 and Phase 2 clinical trials for ulcerative colitis[1][2][6][7][8].
