GLPG2737 is an orally available small-molecule drug developed as a type 2 corrector of the cystic fibrosis transmembrane conductance regulator (CFTR) protein. It was designed to improve the folding and trafficking of mutant CFTR proteins—specifically F508del CFTR—by increasing their membrane expression in combination with other correctors and potentiators. The compound was intended for use in triple combination therapy for cystic fibrosis (CF), aiming to restore healthy activity levels of the defective protein. Additionally, it has been investigated as a CFTR inhibitor for autosomal dominant polycystic kidney disease (ADPKD), where inhibition of CFTR may reduce cyst growth and enlargement. The drug was originally developed by Galapagos NV in collaboration with AbbVie[2][3][4][5][6].
