Overview

GLPG3667 is an investigational, orally administered small molecule that acts as a reversible and selective inhibitor of tyrosine kinase 2 (TYK2). TYK2 is a member of the Janus kinase family involved in cytokine signaling pathways, including type I interferon, interleukin-12/23, and the IL-10 family. By selectively inhibiting TYK2 through ATP-competitive binding, GLPG3667 modulates immune responses implicated in inflammatory and autoimmune diseases. The drug has demonstrated favorable preclinical pharmacokinetics and selectivity for TYK2 over other JAK kinases. In clinical studies, it has shown efficacy signals in moderate-to-severe plaque psoriasis (Phase 1b) and is currently being evaluated in Phase 2 trials for dermatomyositis and systemic lupus erythematosus.
