GLPG3748 (also known as ABBV-3748) is an oral small‑molecule cystic fibrosis transmembrane conductance regulator (CFTR) corrector that emerged from the Galapagos/AbbVie cystic fibrosis modulator collaboration. It is designed to improve the folding and trafficking of mutant CFTR, particularly the F508del variant, thereby increasing functional CFTR protein at the cell surface and enhancing chloride transport in people with cystic fibrosis. GLPG3748 belongs to a family of next‑generation CFTR correctors (including GLPG/ABBV-2222, -2737, -2851, and -3221) that showed robust restoration of CFTR function in F508del-expressing cell systems and was being advanced as part of potential dual or triple combination regimens for cystic fibrosis before the CF program was deprioritized.[3]
