GLPG3970 is a first-in-class, orally available small molecule that acts as a potent and selective dual inhibitor of salt-inducible kinase 2 (SIK2) and salt-inducible kinase 3 (SIK3), with high selectivity over SIK1. It was developed by Galapagos for the treatment of inflammatory diseases, including rheumatoid arthritis, psoriasis, ulcerative colitis, Sjogren's syndrome, systemic lupus erythematosus, and other autoimmune conditions. The drug modulates immune responses by inhibiting pro-inflammatory cytokine production (such as TNFα) while enhancing anti-inflammatory cytokine release (such as IL-10). Preclinical studies demonstrated efficacy in models of arthritis and other inflammatory disorders. Despite promising early clinical activity and biomarker modulation in phase 1/2 trials, development has been discontinued for all indications[1][3][4][5][6].
