Overview

GLPG4399 is a first-in-class, oral, selective inhibitor of salt-inducible kinase 3 (SIK3), developed by Galapagos for the treatment of inflammatory diseases such as rheumatoid arthritis and psoriatic arthritis. It acts by selectively inhibiting SIK3, which leads to reduced production of pro-inflammatory cytokines in various immune cell types including monocytes, macrophages, dendritic cells, B cells, and T lymphocytes. Preclinical studies demonstrated that GLPG4399 reduces disease activity scores and bone erosion in mouse models of arthritis. The drug has shown high selectivity against a panel of 370 kinases and exhibits strong immunomodulatory effects relevant to inflammation[1][3][8].
