Overview

GLPG5101 is an autologous, second-generation anti-CD19/4-1BB chimeric antigen receptor T-cell (CAR-T) therapy developed for the treatment of relapsed or refractory non-Hodgkin lymphoma (R/R NHL). It is manufactured using Galapagos’ decentralized cell therapy platform, which enables rapid production and administration of fresh, stem-like, early memory phenotype CAR-T cells with a median vein-to-vein time of seven days. The product is characterized by an increased proportion of naïve/stem cell memory and central memory CD4+ and CD8+ CAR T cells in the final product compared to starting material. GLPG5101 has demonstrated robust in vivo expansion, durable persistence up to 21 months post-infusion, high antitumor activity across multiple NHL subtypes (including diffuse large B-cell lymphoma, follicular lymphoma, marginal zone lymphoma, and mantle cell lymphoma), and a manageable safety profile with low rates of severe cytokine release syndrome (CRS) and immune effector cell-associated neurotoxicity syndrome (ICANS)[1][2][3][6].
