Overview

GLPG5201 is an autologous chimeric antigen receptor T-cell (CAR-T) therapy under development for the treatment of relapsed or refractory chronic lymphocytic leukemia (CLL), small lymphocytic lymphoma (SLL), and Richter transformation (RT). It is a second-generation CAR-T product candidate that targets CD19 on B cells and incorporates a 4-1BB co-stimulatory domain to enhance T-cell activation and persistence. The therapy involves collecting a patient's own T cells, genetically engineering them to express the anti-CD19/4-1BB CAR construct, and infusing them back as a fresh product following lymphodepleting chemotherapy. The manufacturing process uses point-of-care technology with a median vein-to-vein time of seven days. Clinical studies have shown encouraging safety and efficacy profiles in heavily pretreated patients[2][3][4][8].
