GLR1059 is a next-generation antibody-drug conjugate (ADC) targeting Nectin-4, developed by Gan & Lee Pharmaceuticals. It consists of a novel humanized IgG1 monoclonal antibody site-specifically conjugated to a microtubule-targeting agent payload via a cleavable linker. Unlike the first-generation Nectin-4 ADC enfortumab vedotin, which uses monomethyl auristatin E (MMAE), GLR1059 utilizes a novel mechanism-of-action payload that is not an auristatin derivative. Preclinical studies have demonstrated potent anti-tumor activity in urothelial carcinoma and breast cancer models, including triple-negative breast cancer (TNBC), with a significantly reduced incidence of skin toxicity compared to enfortumab vedotin. The payload is site-specifically conjugated to the antibody via a disulfide bond bridge to generate a homogeneous drug-to-antibody ratio (DAR) of 4.
