This is a four-drug combination regimen consisting of two investigational immuno-oncology monoclonal antibodies, GLS-012 and GLS-010, together with the established chemotherapeutic agents paclitaxel and carboplatin. GLS-012 is an anti-LAG3 monoclonal antibody that inhibits lymphocyte activation gene 3 protein, aiming to enhance T-cell mediated immune responses against tumors[5]. GLS-010, also known as zimberelimab, is a fully human anti-programmed death 1 (PD‑1) monoclonal antibody that blocks the PD‑1 pathway to restore T-cell activity against cancer cells[7]. Paclitaxel is a microtubule inhibitor that disrupts cell division by stabilizing microtubules and preventing their disassembly. Carboplatin is a platinum-based chemotherapy agent that causes DNA crosslinking and apoptosis in rapidly dividing cells[8][6]. The combination of these agents targets both immune checkpoints and tumor cell proliferation pathways. This regimen has been studied in advanced solid tumors after progression on standard treatments, with ongoing phase I/II trials evaluating safety, tolerability, and preliminary efficacy[1][2][3].
