Overview

GLS-5310 is a bi-cistronic DNA vaccine developed for the prevention of COVID-19. It encodes both the SARS-CoV-2 spike (S) protein and the open reading frame 3a (ORF3a) antigen, with ORF3a included as an immunodominant T-cell antigen. The vaccine is administered intradermally followed by application of suction to enhance delivery using the GeneDerm device. Clinical studies have shown that GLS-5310 induces robust antibody and T-cell responses, with high levels of binding antibodies and persistent T-cell immunity observed up to 48 weeks post-vaccination. The mechanism of action involves inhibition of SARS-CoV-2 infection by eliciting immune responses against viral spike glycoprotein and ORF3a antigens[1][3][7].
