Overview

GLSiso2 siRNA is a small interfering RNA (siRNA) designed to selectively silence the expression of the GLSiso2 isoform, also known as GAC (glutaminase C), of the kidney-type glutaminase (GLS) enzyme. Glutaminase is a critical enzyme in cancer metabolism, facilitating glutaminolysis by converting glutamine to glutamate to support the bioenergetic and biosynthetic needs of rapidly proliferating cells. Research presented at AACR 2019 by the University of Sao Paulo demonstrated that GLSiso2 is significantly upregulated in high-grade astrocytomas and glioblastoma compared to normal brain tissue. Silencing this specific isoform using siRNA in U87MG glioblastoma cells resulted in decreased cell proliferation, increased apoptosis, and enhanced sensitivity to the chemotherapeutic agent temozolomide, suggesting its potential as a targeted therapeutic strategy for malignant gliomas.
