Overview
GLT-03 is a novel small-molecule inhibitor of procollagen galactosyltransferase 1 (GLT25D1, also known as COLGALT1), an enzyme that plays a key role in collagen glycosylation and maturation. Identified through AlphaFold2-guided virtual screening, GLT-03 has been shown to suppress hepatic stellate cell (HSC) activation and fibrogenesis. In preclinical studies, treatment with GLT-03 reduced the secretion of procollagen III N-terminal peptide (PIIINP), decreased the expression of fibrogenic markers (such as COL1A1 and alpha-SMA), inhibited HSC migration, and promoted apoptosis, potentially through the modulation of MAPK/ERK and PI3K-Akt signaling pathways. It is being investigated as a potential therapeutic strategy for hepatic fibrosis.
