Drug intelligence / Profile preview
GLT25D1 siRNA is a small interfering RNA (siRNA) therapeutic designed to target and silence the expression of glycosyltransferase 25 domain-containing 1 (GLT25D1), also known as collagen beta(1-O)galactosyltransferase 1 (COLGALT1). GLT25D1 is an enzyme that catalyzes the hydroxylysine O-glycosylation of procollagen, a critical step for collagen triple-helix maturation and secretion. By silencing GLT25D1, the siRNA impairs collagen O-glycosylation, leading to defective procollagen folding, retention within the endoplasmic reticulum (ER), and subsequent ER-associated degradation (ERAD). This structure-oriented approach reduces the secretion of mature collagen and attenuates extracellular matrix remodeling. Developed by researchers at Beijing Ditan Hospital and Capital Medical University, GLT25D1 siRNA (specifically the lead candidate si-D19) is being investigated in preclinical models for the treatment of liver fibrosis.
