Glucagon + GSK1362885 is a combination of two agents with opposing mechanisms relevant to glucose metabolism. Glucagon is a 29-amino acid polypeptide hormone used primarily as an emergency treatment for severe hypoglycemia and as a diagnostic aid in radiologic exams to inhibit gastrointestinal motility. It acts by binding to the hepatic glucagon receptor, stimulating glycogenolysis and gluconeogenesis, thereby increasing blood glucose levels[2][4][5]. GSK1362885 is an investigational small molecule developed by GSK that functions as a potent inhibitor of human glycogen phosphorylase, aiming to reduce hepatic glucose output for the treatment of type 2 diabetes mellitus[6][8]. The combination would theoretically pair the hyperglycemic effect of glucagon with the hepatic glucose output-lowering action of a glycogen phosphorylase inhibitor; however, there are no known approved or clinically developed fixed-dose combinations of these two agents.
