Overview

glucagon-like peptide-2(3-33) is a 31-amino acid peptide metabolite of the intestinotrophic hormone glucagon-like peptide-2 (GLP-2). It is generated endogenously through the proteolytic cleavage of the first two N-terminal amino acids (Histidine and Alanine) of GLP-2(1-33) by the enzyme dipeptidyl peptidase-4 (DPP-4). Unlike the intact hormone, which promotes intestinal mucosal growth and nutrient absorption, GLP-2(3-33) acts primarily as a competitive antagonist at the glucagon-like peptide 2 receptor (GLP2R). In some contexts, it has been observed to display weak partial agonist activity, but its predominant physiological and pharmacological role is the inhibition of GLP-2-mediated signaling. It is extensively utilized as a research tool to investigate the biological actions of the GLP-2 system in gut physiology and diseases such as short bowel syndrome and inflammatory bowel disease.
