Drug intelligence / Profile preview
Glucagon receptor siRNA (GCGR siRNA) is a small interfering RNA (siRNA) therapeutic platform designed to selectively silence the expression of the glucagon receptor (GCGR) in the liver. By targeting GCGR mRNA for degradation, GCGR siRNA inhibits glucagon signaling, which subsequently reduces hepatic glucose production (gluconeogenesis and glycogenolysis) and lowers blood glucose levels. Preclinical studies, particularly those utilizing lipid nanoparticle (LNP) delivery systems, have demonstrated that GCGR siRNA can improve glucose tolerance, reduce HbA1c, and normalize plasma ketone levels in animal models of type 1 and type 2 diabetes. This approach represents a potential gene-silencing strategy for the management of diabetes and associated metabolic disorders.
