Nucien Pharmaceuticals is developing a small molecule glucokinase (GK) activator for the treatment of Type 2 diabetes. Glucokinase serves as the primary glucose sensor in the body, predominantly expressed in pancreatic beta cells and hepatocytes. By allosterically activating GK, this therapeutic candidate enhances the enzyme's affinity for glucose, thereby stimulating glucose-dependent insulin secretion from the pancreas and increasing glucose uptake and glycogen synthesis in the liver. This dual mechanism of action aims to improve glycemic control in patients with Type 2 diabetes. The program is currently in the preclinical stage of development.
