Drug intelligence / Profile preview
GlucoRegulate (also known as tRES-HESP or Glo1 inducer) is a patented, precision dietary supplement formulation combining high-purity trans-resveratrol (tRES) and hesperetin (HESP). Developed by researchers at the University of Warwick and commercialized by GloVitality, the combination acts synergistically to activate the transcription factor Nrf2 (nuclear factor erythroid 2-related factor 2) via its antioxidant response element (ARE) in the GLO1 gene. This activation induces the expression and activity of glyoxalase 1 (Glo1), a key enzyme responsible for detoxifying methylglyoxal (MG), a reactive dicarbonyl glycating agent and byproduct of glycolysis. By reducing MG accumulation and subsequent protein glycation (dicarbonyl stress), GlucoRegulate alleviates the activation of the unfolded protein response (UPR) and downregulates inflammatory mediators such as TNF-alpha, TXNIP, CCL2, IL-8, and RAGE. In clinical trials, the formulation has been shown to improve insulin sensitivity, glycemic control, and vascular function in overweight and obese subjects, making it a potential therapeutic option for the prevention and early-stage management of type 2 diabetes and metabolic dysfunction-associated steatotic liver disease (MASLD).
