Glunektik

Glunektik is a topical gel formulation of the purine nucleoside analog cladribine (2-chlorodeoxyadenosine), developed by the Instytut Matki i Dziecka (Institute of Mother and Child) in Poland. It is specifically designed for the treatment of skin lesions associated with Langerhans cell histiocytosis (LCH) and other histiocytic cell proliferations. As a chemotherapy agent, cladribine acts as a prodrug that is phosphorylated intracellularly into its active triphosphate form, which then inhibits DNA polymerase and ribonucleotide reductase. This mechanism leads to the accumulation of DNA strand breaks and subsequent apoptosis in pathological histiocytes. By utilizing a localized topical delivery system, Glunektik aims to provide effective control of cutaneous manifestations while minimizing the systemic toxicity and side effects typically associated with intravenous or subcutaneous cladribine administration.
