Glunomab is a monoclonal antibody designed to inhibit the specific interaction between tissue plasminogen activator (tPA) and the N-methyl-D-aspartate receptor (NMDAr), particularly the GluN1 (NMDAR1) subunit. In the central nervous system, tPA acts as a neuromodulator that binds to NMDAr, enhancing its activity and potentially driving neuroinflammation and neurodegeneration in conditions like Parkinson's disease (PD). Glunomab blocks this interaction without interfering with the physiological catalytic activity of tPA or the normal ionotropic function of the NMDAr. By preventing tPA-mediated NMDAr overactivation, the antibody reduces microglia activation, immune cell infiltration, and blood-brain barrier permeability. A humanized version of the antibody, LYS241, is being developed by Lys Therapeutics and is currently in IND/CTA-enabling studies for the treatment of Parkinson's disease, with potential applications in other neurological disorders such as multiple sclerosis and stroke.
