Overview

**GLUT5-expressing CAR-T cells** are genetically engineered T cells that ectopically express the GLUT5 fructose transporter alongside a chimeric antigen receptor (CAR), enabling them to utilize fructose as an alternative energy source in glucose-deprived tumor microenvironments (TMEs). This metabolic armoring enhances T-cell proliferation, cytotoxicity, cytokine production, and anti-tumor efficacy both *in vitro* and *in vivo*, particularly against solid tumors and xenografts like PSMA-expressing prostate cancer models (e.g., PC3-LN3-PSMA) or AML, without requiring exogenous fructose supplementation in some cases due to improved glucose competitiveness. Developed through preclinical research, this approach addresses key limitations of standard CAR-T therapies in nutrient-poor TMEs by promoting fructose-driven anaerobic glycolysis and lactate production, maintaining effector functions under repeated antigen stimulation.[1][3][5][7]
