Overview

Glutamine hydroxamate is a synthetic derivative of the amino acid L-glutamine, in which a hydroxamic acid functional group replaces the amide at the γ-position. This modification confers metal-chelating properties and allows it to act as an inhibitor or substrate for various enzymes, particularly those involved in amino acid and carbohydrate metabolism. Its primary mechanism of action is inhibition of enzymes such as glucosamine 6-phosphate synthase (glmS), where it competes with natural substrates, thereby modulating enzyme activity. It is widely used as a biochemical tool to study enzyme kinetics and metabolic pathways, especially in research on nitrogen metabolism and hexosamine biosynthesis. Glutamine hydroxamate has not been approved for clinical use but serves as an experimental agent in preclinical studies[1][2][3][4].
