Overview

**Glutathione** is an endogenous reduced tripeptide antioxidant composed of glutamate, cysteine, and glycine. Administered glutathione replenishes thiol-based cellular redox capacity and serves as a substrate for glutathione peroxidases and glutathione S-transferases, supporting detoxification of reactive oxygen species, peroxides, and electrophilic metabolites. It has been used or studied as a cytoprotective adjunct through oral, intravenous, inhaled, and intranasal delivery, including for chemotherapy-associated toxicities, cystic fibrosis, and Parkinson's disease. Clinical evidence for disease-modifying or symptomatic benefit remains limited and route-specific. A 2024 retrospective breast-cancer analysis associated prolonged oral glutathione exposure during chemotherapy with poorer outcomes, so use during cytotoxic chemotherapy requires particular clinical caution.
