Overview

GLV-1h153 is an engineered oncolytic vaccinia virus developed by Genelux Corporation, derived from the GLV-1h68 (Lister strain) backbone. It is specifically designed for both therapeutic oncolysis and deep-tissue imaging by expressing the human sodium iodide symporter (hNIS) and green fluorescent protein (GFP). The hNIS transgene allows for the concentration of iodine isotopes (such as 124I or 131I) within infected tumor cells, facilitating non-invasive monitoring via PET/SPECT imaging and potentially enabling targeted radiotherapy. In preclinical models of pancreatic adenocarcinoma, GLV-1h153 has demonstrated the ability to selectively replicate in cancer cells, induce immunogenic cell death, and alter gene expression profiles related to DNA repair and anti-apoptotic signaling, thereby overcoming resistance to conventional therapies.
