GLV-1h43 is an engineered oncolytic vaccinia virus (VACV) derived from the Lister strain, developed by Genelux Corporation. It is a derivative of the GLV-1h68 (GL-ONC1) backbone, specifically modified to express the human sodium iodide symporter (hNIS) gene. This modification enables a theranostic approach to cancer treatment: the virus selectively replicates in and lyses tumor cells (oncolysis) while the expression of hNIS allows for non-invasive, real-time monitoring of viral distribution and replication using PET or SPECT imaging with radiotracers like I-124 or Tc-99m. Furthermore, the hNIS expression facilitates the targeted uptake of therapeutic radioisotopes, such as I-131, into infected tumor cells, thereby combining viral oncolysis with localized radiotherapy to enhance anti-tumor efficacy.
