GLV8 is a novel synthetic flavonoid being investigated for the treatment of venetoclax-resistant acute myeloid leukemia (AML). It functions by targeting and down-regulating the expression of G protein-coupled receptor 84 (GPR84), a pro-inflammatory receptor that responds to medium-chain fatty acids and has been identified as a potential oncogenic gene in AML. In preclinical studies, GLV8 has demonstrated the ability to inhibit cell proliferation, suppress colony formation, and induce apoptosis in both venetoclax-sensitive and resistant AML cell lines. This activity is associated with the dose-dependent downregulation of the anti-apoptotic proteins MCL1 and BCL2, suggesting that GLV8 may overcome resistance mechanisms in leukemia patients who have failed standard venetoclax-based therapies.
