Overview

GLWL-01 is a selective, reversible inhibitor of ghrelin O-acyltransferase (GOAT), the enzyme responsible for converting unacylated ghrelin (UAG) into acylated ghrelin (AG). AG stimulates appetite and is elevated in conditions such as Prader-Willi syndrome. GLWL-01 was developed to treat hyperphagia in Prader-Willi syndrome and has also been investigated for alcohol use disorder and type 2 diabetes mellitus. The drug is administered orally and has demonstrated safety and tolerability in clinical trials, with pharmacological evidence confirming GOAT inhibition. However, it did not significantly reduce hyperphagia-related behavior or improve clinical endpoints related to obesity or metabolic parameters[6][7][4].
