Drug intelligence / Profile preview
GLY-202 (synthetic heparan sulfate 18-mer) is a structurally homogeneous, non-anticoagulant synthetic heparan sulfate octadecasaccharide being developed by Glycan Therapeutics in collaboration with the University of North Carolina at Chapel Hill. It is designed as an anti-inflammatory agent for the treatment of sepsis and acute liver injury, including acetaminophen-induced liver failure. Unlike natural heparin, GLY-202 is engineered to lack anticoagulant activity, thereby avoiding bleeding risks. It acts by binding directly to and neutralizing extracellular histone H3 and high mobility group box 1 (HMGB1), which are key damage-associated molecular patterns (DAMPs) that drive sterile inflammation and tissue damage. By blocking the HMGB1/RAGE pathway and neutralizing histones, GLY-202 reduces neutrophil infiltration, inhibits pathological coagulation activation, and prevents liver and kidney injury.
