Gly-low is a multi-ingredient dietary supplement cocktail developed by the University of California, San Francisco (UCSF) in collaboration with the Buck Institute for Research on Aging. It is composed of five FDA-recognized Generally Regarded As Safe (GRAS) compounds: alpha-lipoic acid, nicotinamide (vitamin B3), thiamine (vitamin B1), pyridoxamine (vitamin B6), and piperine. The formulation is designed to lower levels of methylglyoxal (MGO), a reactive byproduct of glycolysis, thereby reducing the formation of advanced glycation end-products (AGEs). By targeting the glycation pathway, Gly-low aims to mitigate age-related metabolic dysfunction, reduce food intake by modulating hypothalamic ghrelin signaling, and improve insulin sensitivity. It is currently being evaluated in Phase 1/2 clinical trials for the treatment of obesity and age-related conditions such as frailty and cognitive decline.
