Overview

Glycocholate is a glycine-conjugated bile acid being investigated by Västra Götalandsregionen for its potential to improve clinical outcomes in patients following Roux-en-Y gastric bypass (RYGB). In RYGB patients, the bypass of the proximal small intestine and the asynchronous delivery of bile acids to the distal gut often result in impaired micellar solubilization of fats and minerals, leading to significant calcium malabsorption and subsequent bone metabolism disorders such as secondary hyperparathyroidism. Oral administration of glycocholate is intended to restore effective micelle formation in the distal intestine, thereby enhancing the bioavailability of calcium and fat-soluble vitamins. Beyond its physical-chemical role in digestion, glycocholate also acts as a signaling molecule, serving as an agonist for the farnesoid X receptor (FXR) and the G protein-coupled bile acid receptor 1 (TGR5), which regulate metabolic homeostasis.
