GlycoConnect bispecific ADCs (bsADCs) represent a next-generation class of antibody-drug conjugates developed using Synaffix's (a Lonza company) site-specific conjugation platform. This technology targets the native N-linked glycans of bispecific antibodies to attach cytotoxic payloads, ensuring a highly homogeneous product with a precise drug-to-antibody ratio (DAR). By utilizing a bispecific antibody format, these ADCs can simultaneously target two different tumor-associated antigens (TAAs) or two distinct epitopes on the same antigen (biparatopic), potentially overcoming resistance mechanisms and improving tumor selectivity. The platform typically incorporates the HydraSpace® highly polar spacer to improve the therapeutic index and stability, along with payloads from the toxSYN® platform. Preclinical studies have demonstrated that GlycoConnect bsADCs offer superior homogeneity, stability, and in vivo antitumor activity compared to traditional randomly conjugated bispecific ADCs.
