Glycodeoxycholic acid (gDCA) is a secondary bile acid, specifically the glycine-conjugated form of deoxycholic acid, currently under investigation as a potential treatment for type 2 diabetes mellitus and other cardiometabolic disorders. It functions as a small molecule agonist of the Takeda G protein-coupled receptor 5 (TGR5) and the Farnesoid X receptor (FXR). By activating these pathways, gDCA influences lipid, glucose, and energy metabolism, including the stimulation of fibroblast growth factor 19 (FGF19) production. Research is primarily led by the Academic Medical Center (AMC) at the Amsterdam University Medical Centres, with clinical studies evaluating oral formulations, including enteric-coated capsules for targeted release, both as monotherapy and in combination with ezetimibe.
