Glycomimetic 153 is a synthetic glycomimetic ligand designed to target and block the Siglec-9 receptor, an inhibitory checkpoint on immune cells such as Natural Killer (NK) cells. Siglec-9 typically binds to sialic acid-containing glycans, such as those found on the mucin MUC16, which is overexpressed in ovarian cancer cells. By blocking this interaction, glycomimetic 153 prevents the inhibitory signaling that allows cancer cells to evade the immune system, thereby enhancing the cytolytic activity and recognition of tumor cells by NK cells. Developed by researchers at the University of Wisconsin-Madison, it is being investigated for its potential to augment antitumor immunity in epithelial ovarian cancer.
