The glycosylated MUC1 peptide vaccine represents a class of investigational cancer immunotherapies designed to target Mucin-1 (MUC1), a transmembrane glycoprotein that is highly overexpressed and aberrantly O-glycosylated in various adenocarcinomas, including breast, ovarian, and pancreatic cancers. Unlike non-glycosylated MUC1 vaccines, which often fail to mimic the native conformation of tumor-associated MUC1, these vaccines utilize synthetic glycopeptides derived from the MUC1 tandem repeat region. These glycopeptides bear tumor-associated carbohydrate antigens (such as Tn or TF antigens) to stimulate both humoral (antibody-mediated) and cellular (T-cell) immune responses specifically against cancer cells while sparing normal tissues. A notable clinical-stage candidate within this platform is ONT-10, developed by Oncothyreon (later Cascadian Therapeutics), which entered Phase I clinical trials.
