GLYN441 is a preclinical small molecule drug candidate developed by NeuroBio.ai in partnership with Galyan Bio. It is designed to target the alpha-7 nicotinic acetylcholine receptor (α7nAChR) to treat Alzheimer's disease and glioblastoma. The drug's mechanism of action involves blocking the interaction between the α7nAChR and T14, a neurotoxic 14-amino acid peptide derived from the C-terminus of acetylcholinesterase (AChE). In Alzheimer's disease, T14 binding to an allosteric site on the α7nAChR triggers excessive calcium influx, leading to excitotoxicity and neuronal death; in glioblastoma, this same pathway is implicated in tumor cell proliferation. By acting as a negative allosteric modulator of this interaction, GLYN441 aims to provide a disease-modifying approach for neurodegeneration and a novel therapeutic strategy for brain cancer.
