GlyphAllo is a novel oral prodrug of allopregnanolone in clinical development for major depressive disorder (MDD) with or without anxious distress. It is designed to overcome the poor oral bioavailability of allopregnanolone, which is an endogenous neurosteroid with antidepressant, anxiolytic, and sleep-promoting effects. GlyphAllo achieves high oral exposure, robustly engages its CNS target—as shown by changes in EEG beta power and saccadic eye velocity—and regulates the hypothalamic-pituitary-adrenal axis as indicated by cortisol reduction under stress in Phase 2a studies. Developed as a first-in-class treatment platform, GlyphAllo is being evaluated in a Phase 2b randomized, double-blind, placebo-controlled trial (BUOY-1) in adults with MDD, with or without anxious distress. The drug acts by engaging the γ-aminobutyric acid type A (GABA-A) receptor, mimicking the endogenous effect of allopregnanolone to dampen stress and improve depressive and anxious symptoms[1][3][5][7][9].
