GlyTR1 CAR T cells are an experimental chimeric antigen receptor (CAR) T-cell therapy designed to target Tumor Associated Carbohydrate Antigens (TACAs), specifically β1,6GlcNAc-branched N-glycans. Developed using the Glycan-dependent T cell Recruiter (GlyTR) technology, these cells utilize high-avidity, velcro-like lectin binding rather than traditional high-affinity antibody binding. This mechanism allows the CAR T cells to selectively kill cells with high TACA density (typical of cancer cells) while sparing normal tissues with low TACA expression, thereby reducing on-target, off-cancer toxicity. The project is supported by the NCI Cancer Moonshot program and has shown pan-cancer activity in preclinical models.
