GlyTR2 is a biologically engineered bispecific protein developed as part of the Glycan-dependent T cell recruiter (GlyTR) platform, designed to treat multiple types of cancer. The drug targets tumor-associated carbohydrate antigens (TACAs), specifically those with high density on malignant cells but low expression or absence in normal tissue, such as the Tn antigen. GlyTR2 combines a carbohydrate-recognition domain (CRD) from a lectin for glycan targeting with a single-chain variable fragment (scFv) from an antibody targeting CD3 on T cells, thus recruiting cytotoxic T cells directly to tumor sites. Its "velcro-like" high-avidity binding specifically accumulates GlyTR2 at tumors due to their high TACA density, leading to potent T cell-mediated killing of cancer cells without significant harm to healthy tissue. GlyTR2 has demonstrated preclinical efficacy against breast, ovarian, colon, pancreatic, lung, prostate cancers, and leukemia, with broad pan-cancer activity. Notably, GlyTR2 is also designed to overcome tumor-induced immunosuppression, a major limitation in current immunotherapies. Clinical-grade manufacturing is underway, and the drug is advancing toward first-in-human clinical trials for metastatic solid tumors, with Phase 1 trials expected to initiate within approximately two years[1][3][5].
