Overview

GM-0111 is a synthetic glycosaminoglycan (GAG) being developed as a prophylactic agent for radiation-induced proctitis (RIP). It exerts radioprotective effects through a multi-faceted mechanism involving the inhibition of C-C motif chemokine ligand 5 (CCL5), a potent chemotactic agent for immune cells. GM-0111 binds directly to CCL5 with high affinity (Kd ~3.34 nM), inhibiting its ability to induce mast cell chemotaxis and degranulation, and also reduces CCL5 secretion from endothelial cells. Additionally, it attenuates oxidative stress by decreasing reactive oxygen species (ROS) production. Preclinical studies in murine models have demonstrated its potential to mitigate tissue damage and inflammatory responses resulting from ionizing radiation.
