Overview

GM-10395 is an orally available, selective small-molecule inhibitor of pyruvate dehydrogenase kinase 4 (PDK4), developed by JD Bioscience in collaboration with Kyungpook National University. PDK4 is a mitochondrial enzyme that regulates glucose oxidation and lipid metabolism by inhibiting the pyruvate dehydrogenase complex (PDC). By selectively inhibiting PDK4, GM-10395 restores PDC activity, enhances mitochondrial respiration, reduces reactive oxygen species (ROS) generation, and improves insulin-responsive AKT phosphorylation. Preclinical studies have demonstrated its therapeutic potential across multiple indications, including type 2 diabetes mellitus, insulin resistance, hepatic steatosis, obesity, inflammatory bowel disease (colitis), age-related macular degeneration (choroidal neovascularization), and acute kidney injury (ischemia-reperfusion injury).
