The GM-CSF-secreting breast cancer vaccine is a therapeutic cancer vaccine composed of genetically modified human breast cancer cells engineered to secrete granulocyte-macrophage colony-stimulating factor (GM-CSF). There are both autologous and allogeneic versions. In the autologous approach, tumor cells are harvested from the patient, transduced with a replication-defective adenoviral vector encoding GM-CSF, irradiated to prevent proliferation, and then administered back to the patient as a personalized immunotherapy. The allogeneic version uses established cell lines similarly engineered to secrete GM-CSF. Upon administration (typically intradermal), these vaccines aim to stimulate an anti-tumor immune response by recruiting and activating antigen-presenting cells at the site of injection via secreted GM-CSF, thereby potentiating tumor-specific T cell responses against breast cancer antigens[3][5][6]. The primary indication is for patients with metastatic or stage II-III breast cancers.
