A GM-CSF-secreting tumor vaccine is a **personalized or allogeneic cancer vaccine** in which tumor cells (either autologous or allogeneic) are genetically engineered to secrete granulocyte-macrophage colony-stimulating factor (GM-CSF). Upon administration, these cells secrete GM-CSF locally, stimulating the recruitment and activation of dendritic cells (DCs) at the site of injection. The DCs then process and present tumor-associated antigens (TAAs) from the vaccine cells, leading to the priming and activation of tumor-specific CD4+ and CD8+ T cells, thereby inducing an anti-tumor immune response. This vaccine does not require HLA matching between the patient and the vaccine cells. GM-CSF-secreting tumor vaccines have been explored in multiple solid tumors including melanoma, prostate cancer, renal cell carcinoma, pancreatic cancer, breast cancer, and non-small cell lung cancer. Development has included both autologous (patient-derived cells) and allogeneic (cell lines or donor-derived cells) formulations[1][2].
