The GM-K562 and autologous glioma cell vaccine is an experimental cellular immunotherapy developed by Massachusetts General Hospital for the treatment of recurrent malignant glioma. This therapeutic vaccine is a variation of the GVAX platform, utilizing a combination of two cell types: lethally irradiated autologous tumor cells and lethally irradiated GM-K562 cells. The autologous cells provide a personalized source of the patient's own tumor-associated antigens. The GM-K562 cells are an allogeneic bystander cell line genetically engineered to continuously secrete Granulocyte-Macrophage Colony-Stimulating Factor (GM-CSF). Upon administration, the locally produced GM-CSF acts as a potent immune adjuvant, recruiting and activating professional antigen-presenting cells, such as dendritic cells, to the injection site. these cells then process the tumor antigens and migrate to regional lymph nodes to prime a systemic T-lymphocyte and humoral immune response against the glioma.
